(increases, E2F1 Activity)
Evidence: Sp1 and E2F binding sites are both essential for activation of the murine thymidine kinase promoter (Karlseder et al., 1996). The promoter was activated when the Sp1 and E2F sites were separated by 6 or 10 bp but not when they were separated by 20 bp. The DHFR promoter was strongly activated by Sp1 alone but hardly at all by E2F1 alone (cotransfection in insect cells) (Lin et al., 1996a). E2F1, however, enhanced the activating ability of Sp1, even in the absence of a functional E2F binding...

(Breast Neoplasms) :
Transcriptional activation of
E2F1 gene expression by 17beta-estradiol in MCF-7 cells is
regulated by
NF-Y-Sp1/estrogen receptor interactions ... Thus, transcriptional activation of
E2F1 gene expression in MCF-7 cells by E2 is
regulated by multiprotein
ER/Sp1-NF-Y interactions at GC-rich and two CCAAT elements in the proximal region of the E2F1 gene promoter
Nicolás et al., Biochem J 2003
:
Whereas overexpression of NF-Y or
E2F had an additive effect on
Sp1 overexpression, the activation of Sp1 transcription due to Sp1 was counteracted by Sp3 overexpression
Tapias et al., Cell cycle (Georgetown, Tex.) 2008
:
Among the known Sp1 interacting proteins,
E2F-DP1 , Cyclin D1, Stat3 and Rb
activated the
Sp1 promoter, whereas p53 and NF kappaB inhibited it
Hazar-Rethinam et al., J Invest Dermatol 2011
:
Transient transfection of E2F1-E2F6 indicated that
E2F mediated reduction of
Sp1 transcription was not responsible for E2F mediated suppression of squamous differentiation
Shao et al., Oncogene 1995
:
Since Rb is a regulator of E2F and Sp1 mediated transcription, we have examined the effect of overexpression of cyclins on transcription mediated by
E2F dependent adenovirus E2 promoter and by
Sp1 in a cotransfection assay in 3T3 cells
